A 14-year-old girl, was attacked by two men on the upper deck shortly after getting on board at Silverburn with another 14-year-old girl shortly after midnight.
The ordeal came to an end when her companion persuaded passengers sitting downstairs to go up and intervene.
The driver ordered the attackers to leave the bus as it passed through Shawlands at about 12.30am.
Police said the two 14-year-old girls initially sat downstairs, before the victim went to the upper deck and spoke with two men who were there.
A police spokesman said: “She was assaulted by both of the men and then seriously sexually assaulted by them.
“Her friend then went upstairs to see her and raised the alarm with some of the other passengers downstairs.
“Two men and a woman from the bus assisted the girl and her friend, and they left the bus two stops from Silverburn and made their way home on another bus. Police were then contacted.”
Both suspects are described as 18 to 20 years old. One of them had a shaved head, was around 5ft 10ins and was wearing a black jumper.
The second man is described as having brown spiky hair, wearing a tracksuit which may have been grey, with a body warmer or jacket.
