Cops arrested Elizabeth Garner, a former cheerleader for the Tennessee Titans, after she was accused of the offense.
Garner followed the 12-year-old boy into a bathroom during his parents' house party, police have said.
The boy's mom called police after he told her what had happened.
Garner, 42, is a former cheerleader for the Tennessee Titans until 2009.
The married blonde claims innocence.
She told investigators she was "drunk that evening and she got the boy confused with a man" also at the party.
Garner was released on $30,000 bail and faces more than eight years in prison if convicted.
But local police say that's "no excuse for sexually assaulting a child."
Murfreesboro police officer Kyle Evans said: "If you can't tell the difference between a 12-year-old and an adult, then there are problems."
